Introduction into Microscope Activity

Introduction:

Microscopes are very important tools that we use in Biology. The term Microscope is basically
referred to as “viewing the tiny” because the microscopes that we use are used to study things
that can be too small and can be observed by using other methods. The type of Microscope that
had been used in this experiment was a compound light microscope. The term compound means
that our microscope passes through the specimen using light and lenses. When using a
microscope like this, we study specimen on a glass slide.

In this lab, we learned the parts of a microscope and its functions, how to calculate the
magnification of a compound light microscope, how to make a wet mount slide, and how to
orientate and make movement of the specimens image changes when they are viewed through a
compound light microscope. Additionally, we had two different lens systems for magnification,
ocular lens, and the objective. Ocular lens has a magnification of 10X. These microscopes can
have 4 objectives attached to the nosepiece located above the stage.

Materials:

Materials used were our compound light microscopes set up in the lab, slides, cover slips, and
lens paper. To start out the procedure, as in our lab groups, we plugged in our microscope. We
cleaned out our oculars, objectives, and the condenser, using lens paper dampened on one end
with lens cleanser. Objectives had been cleaned from lowest to highest power. Adjusting the eye
width of the oculars, first step we started off with was gently taking a slide flat on the stage and
pushing it up into the clamp until our edges of the slide were meeting at the back of the clamp.
Centering the specimen over the light source, when using the 4X and 10X objectives: the coarse
and adjustment knobs were used. When we used the 4x and 10x objectives, we had only been
