IMMUNOLOGIE PRACTICUMTOETS SAMENVATTINGDendritische cellen opkweken voor immunohherppie
Doel: het genereren van dendritishe sellen (DCi) uit humaan bloed die Th-1 sel astvate eunnen
induseren en eunnen migreren naar lymfoïde organen. De DCi zijn op deze manier zo geishiet
mogelije voor immunotherapie omdat Th-1 sellen sytotoxiishe T-sellen hulp bieden bij bijvoorbeeld
het beitrijden van tumorsellen.
Mehhode: Monosyten zuiveren, vervolgeni diferentate tot
imDCi. Zei verishillende maturatemixen werden gebruiet op
deze imDCi, waarna een IL-6 en IL-12 ELISA voor
sytoeineproduste-metng en een migrate-isoring werden
uitgevoerd.
1. Monocyhen isoleren uih bufycoph
Ssheiden d.m.v. opbrengen op fsoll, sentrifugeren, ringfraste
afnemen, waiien, sentrifugeren, afgieten ets. en daarna te
isheiden met een magneet (MACS).
- Bufycoph: bevat 20% monosyten, dit breng je op fsoll op om het
te isheiden. De dishtheid van Fisoll zit presiei tuiien PBMCi en
erytrosyten met granulosyten in.
- TNC: antitollingimiddel zodat plaima geen ierum wordt,
itollingifastoren blijven intast
- Cellen hellen: 25 hoejei ii 0,1misroliter. 1:2 verdunnen met
trypaanblauw
- MACS-bufer: Bevat EDTA, verlaagt seladheiie waardoor de sellen
dui niet plaeeen aan je materiaal.
- CD14 microbepds (MACS): deze worden geeoppeld aan CD14 op
de monosyten en worden geisheiden in een MACS (Magnets sell
ieparator), een eolom met magneet eraan. De misrobeadi blijven
hangen door de magneet. Door de magneet weg te halen en
daarna met Cellgro-medium en de zuiger door te duwen eonden de
monosyten gezuiverd worden.
2. Monocyhen nppr imDCs
IL-4 en GM-CSF (itmuli) toevoegen aan seliuipeniiei in medium.
Insubate van 6 dagen bij 37 graden. Diferentate wai te zien aan
de groote van de sellen.
3. imDCs nppr mDCs
DCi matureren met PAMPi en sytoeinei. Dit verhoogt sytoeine en
so-itmulatoir molesuul expreiiie, maar ean ooe de
migratesapasiteit verhogen. Mature DCi erijgen uitloperi
waarmee ze eunnen migreren.
- Mphurptiemixen: FCS (foetaal ealfierum), PAMPi (MPLA, LPS) en
sytoeinei (IFN-gamma, TNF-alpha, IL-1-Beta, PGE 2).
