What is it?
CRISPR- Clustered Regularly Interspaced Short Palindromic Repeats
Cas- CRISPR associated systems
• CRISPR method is based on a natural system in bacteria to protect themselves
from virus infections.
• When a virus infects a bacteria, the bacteria produces two strands of short RNA,
one of which contains a sequence that matches that of the invading virus.
• The two strands form a complex together with the Cas9 protein, which is a
nuclease (cuts DNA).
• When the matching sequence (known as the guide RNA) finds the target region
of the viral DNA, the Cas 9, cuts the viral DNA, thus disabling the virus.
• The complex locks at a sequence (PAM) in the genome.
• The Cas9 unzips the DNA and matches it to its target RNA (gude RNA).
• If the match is correct, Cas9 cuts the DNA (both strands)
