Abstract

The experiments and procedures outlined in this report discuss the effects of lactose

intolerance which affects millions of people worldwide. The disaccharide Lactose is made

of up the monomers beta-D-galactose and beta-D-glucose. It is an enzyme that is found in

various organs throughout the bodies of mammals, and is used as an energy source in cells.

There have been many advances in the in the production of enzymes that help individuals

who are affected by lactose intolerance. These products assist people who lack these

enzymes in breaking down Lactose. The purpose of this laboratory experiment is to test

Lactose for the chemical properties of the enzyme, and to arrive at a conclusion as to the

determination of if the enzyme Lactase came from human or bacterial cells. The first

experiment involves the testing of the effects of temperature on enzyme activity, by

exposing the enzyme to different set levels of temperature and recording the effect. It is

hypothesized that the enzyme will follow denaturation, in that it will be effective to a

certain extent until the temperature becomes too high and begins to denature. The second

experiment encompasses the effects of pH on enzyme activity to discover the effects that

different pH levels have on enzyme activity. It is hypothesized that the effects will be

similar to the effects of denaturization as well, working effectively to an extent to when the

pH is either too high or too low will begin to denature. The third experiment will specify if

the enzyme lactase will be most likely to interact or bind with lactose compared to maltose.

The final experiment will determine what the specific cofactors of the enzyme lactase are,

as well as the role that metal ions will have on the activity of lactase.
