you. This ELN is due Wednesday October 30 th no later than 11 am in the appropriate assignment folder
on A2L. Please submit one pdf file of this completed template. Please note: it is your responsibility to
ensure that your submitted files are not corrupt and can be properly opened or your submission will be
incomplete. Additionally, you must submit in the appropriate assignment folder on A2L otherwise your
submission will be incomplete.
Purpose and experimental overview
In the space provided below:
1. Purpose - Briefly describe the purpose of the lab. What is your overall goal for this lab day
(very concise, cannot exceed 200 words)?
The purpose of this lab is to isolate the E.coli DH5 cells and expose them to alkaline lysis in order
to obtain the plasmid DNA containing the folA gene. The overall goal for this lab is to gain
experience in reading, understanding, and extracting vital information from a kit manual.
2. Experimental overview – Please create a flowchart (using software such as PowerPoint) to highlight
the miniprep procedure (please include diagrams, safety considerations, brief protocol, incubation
times, centrifugation speeds, etc.). The flowchart must be your original work and cannot be copied
directly from the PureLink manual (please note: you will be purifying your plasmid DNA using a
centrifuge). Please also include the cell growth (page 80) and lysis procedure prior to the actual
DNA purification procedure. Also, page 83 of your courseware has some specific requirements for
the miniprep procedure. Please include this information in your flowchart.
