In preparation for the lab day, please read the lab in its entirety and fill out the following sections.
Thank you. This ELN is due Wednesday Oct 23 rd no later than 11 am in the appropriate assignment
folder on A2L. Please submit one pdf file of this completed template. Please note: it is your
responsibility to ensure that your submitted files are not corrupt and can be properly opened or your
submission will be incomplete. Additionally, you must submit in the appropriate assignment folder on
A2L otherwise your submission will be incomplete.

Purpose and experimental overview
In the space provided below:

1. Purpose - Briefly describe the purpose of the lab. What is your overall goal for this lab day (very
concise, cannot exceed 250 words)?

To isolate and manipulate DNA through DNA purifications and DNA ligation. DNA will be purified from
the 2 restriction digestion reactions using the PureLink TM Quick Gel Extraction and PCR Purification
Combo Kit. Ligation reactions of folA gene and digested/ purified pET26b will be set up. Using heat
shock transformation, ligation reaction products will then be introduced into E. coli DH5 bacteria.
The transformed cells will be plated on LB agar plates containing the appropriate antibiotic.




2. Experimental overview – Please create a flowchart (using software such as PowerPoint) to
highlight only the PCR purification procedure using the kit specified in the courseware (please
include diagrams, safety considerations, brief protocol, incubation times, centrifugation speeds,
etc.). The flowchart must be your original work and cannot be copied directly from the PureLink
manual.
